Glutathione Peptide Overview

Glutathione (GSH) is a naturally occurring tripeptide of glutamate, cysteine and glycine, joined by an unusual gamma peptide bond that makes it resistant to ordinary peptidase activity. It is present in nearly every mammalian cell, often at millimolar concentrations, and is the most abundant low-molecular-weight thiol in the body. In research settings it is the reference compound for studying cellular redox balance.

The Cell’s Primary Antioxidant

The cysteine thiol group allows glutathione to donate an electron directly to reactive oxygen species, neutralising them and forming the oxidised dimer GSSG. The ratio of reduced to oxidised glutathione is widely used as a quantitative readout of a cell’s redox state, and depletion models are a standard way to induce controlled oxidative stress.

Enzymatic Cofactor and Redox Cycling

Glutathione is the required cofactor for glutathione peroxidase, which reduces hydrogen peroxide and lipid peroxides, and for the glutaredoxin system, which reverses protein S-glutathionylation. Glutathione reductase then regenerates the reduced form using NADPH, creating a continuous cycle that research models use to study antioxidant capacity under load.

Phase II Detoxification

Through the glutathione S-transferase family, glutathione conjugates to electrophilic xenobiotics and reactive metabolites, marking them for export and elimination. This makes it central to hepatotoxicity models, where glutathione depletion is a well-characterised marker of chemical liver injury.

Immune and Inflammation Research

Intracellular glutathione levels in antigen-presenting cells have been shown in research models to shift cytokine response patterns, and the compound is studied for its role in lymphocyte proliferation and redox-sensitive inflammatory signalling.
